New Paper: Unraveling protein and gene interactions involved in HIV research
Irrinki et al. investigated the use of structure-based drug design to create novel Protein kinase C (PKC) agonists for activation of latent HIV reservoirs, which help improve recognition and elimination of HIV-infected cells. (DOI: 10.1371/journal.ppat.1012874)
The group designed agonists with the goal to selectivity bind PKC- ε, reduce platelet toxicity, and strongly target latent HIV expression.The researchers carried out multiple experiments, including xMAP-based assays for protein and gene expression analysis. A custom Invitrogen™ QuantiGene™ Plex multiplex panel was designed to measure EGR1 and CD69 mRNA in rat blood throughout different timepoints after PKC agonist dosing. The Invitrogen™ ProcartaPlex™ NHP Cytokine/Chemokine/Growth Factor Panel, 37plex was used for plasma cytokine analysis of PKC agonist-dosed rhesus macaques across selected time points.
With the use of different protein and gene expression assays, the group was able to show that selective PKC isoform activation may open new avenues towards HIV therapeutic strategies.
